适应症
毒蕈碱受体拮抗剂Enablex是为膀胱过动症与急迫性尿失禁，紧迫性和频率症状的治疗表示。
剂量和用法
建议的出发Enablex缓释片剂量为7.5毫克，每日一次。根据个人的反馈表明，剂量可增至15毫克，每天一次，如早两个星期后开始治疗。
对Enablex每日剂量不应超过7.5毫克以下患者：
中度肝功能不全患者（Child - Pugh分级乙）
患者服用烈性CYP3A4抑制剂
Enablex不建议病人使用严重肝受损（Child - Pugh分级丙）。
Enablex可采取或没有食物。吞下的药片应整水，而不是咀嚼，分裂或挤压。

缓释片7.5毫克和15毫克。
禁忌
Enablex是禁忌的患者，或在风险中，下列条件：
尿潴留，
胃潴留，或
失控的窄角型青光眼。
注意事项：

不良反应
最常见的不良反应为Enablex（“3％）是：便秘，口干，头痛，消化不良，恶心，尿路感染，意外受伤，和流感症状。
药物相互作用
注意事项时，应采取Enablex是随之而来的是主要由CYP2D6和代谢有一个狭窄的治疗窗口，如氟卡尼，甲硫达嗪和三环类抗抑郁药，药物使用。
与其他的Enablex抗胆碱药物同时使用可能会增加的频率和/口干，便秘或严重程度，视力模糊等抗胆碱药理作用。抗胆碱药物可能会潜在地改变一些随之而来给药因肠胃蠕动影响吸收。
利用在种群
怀孕：Enablex应该在怀孕期间使用只有在对母亲的潜在利益大于对胎儿的风险。

达非那新(darifenacin，Enablex)治疗膀胱过动症
随着我国人口老龄化问题的日益严重，人们生活水平的提高，针对老年人群疾病治疗药物的开发也日益受到药学工作者的重视。
诺华制药公司（Novartis）日前宣布，FDA批准其达非那新（darifenacin，Enablex）7.5和15mg缓释片剂用于治疗伴有急迫性尿失禁、尿急、尿频症状的膀胱过动症（OAB）患者。本品一日1次给药，预计2005年早期在美国上市。
M3受体主导膀胱逼尿肌的收缩，本品即通过阻断M3受体来发挥作用。本品系一种有效的毒蕈碱性受体抑制剂，有助于减少尿失禁发作、增加膀胱储尿量、减少尿频次数、降低尿急相关的排尿压力和急迫性。
FDA此次批准基于4项关键性临床研究提供的疗效数据和多项研究提供的安全性数据。研究共纳入7000余例平均年龄为58岁的患者，患者采用多种剂量的本品治疗。在这些研究中，本品组患者的尿失禁次数和排尿次数均取得减少，膀胱容量增大，尿急感降低。本品使每周的尿失禁次数减少了83%。本品在治疗头2周即显效，在为期12周的临床研究期间疗效恒定。本品的长期安全性临床研究一直持续了1年。
迄今为止，本品已进行过98项临床研究，纳入过1万余例患者。在临床研究中，本品最常见的不良反应有口干、便秘，但因此导致的患者停药率很低。本品的不良反应多为轻至中度，多发于治疗的头2周。与其它的OAB治疗药物同样，尿潴留、胃潴留、难治性闭角型青光眼患者或处于上述疾病风险中的患者不宜使用本品。

Enablex (Darifenacin) Extended Release Tablet is an anticholinergic used to treat overactive bladder in patients who frequently experience symptoms such as an urgent need to urinate, a need to urinate often, or leakage of urine when there is an urgent need to urinate. Enablex (Darifenacin) Extended Release Tablets may also be used to treat other conditions as determined by your doctor.
ENABLEX SIDE EFFECTS that may occur while taking Enablex (Darifenacin) Extended Release Tablets include dizziness, constipation, dry mouth, dry eyes, blurred vision, decreased amount of urine produced, stomach upset or pain, nausea, or weakness. If they continue or are bothersome, check with your doctor.
CHECK WITH YOUR DOCTOR AS SOON AS POSSIBLE if you experience an unusual or severe headache, back pain, pain below the ribs, fever, chills, bloody or cloudy urine, painful urination, or if you are unable to urinate despite an urge to do so.
An allergic reaction to Enablex (Darifenacin) Extended Release Tablets is unlikely, but seek immediate medical attention if it occurs.
Symptoms of an allergic reaction to Enablex (Darifenacin) Extended Release Tablets include rash, itching, swelling, severe dizziness, or trouble breathing. If you notice other Enablex side effects not listed above, contact your doctor, nurse, or pharmacist.
You must swallow the Enablex (Darifenacin) Extended Release Tablets whole. Do not break, crush, or chew the Enablex (Darifenacin) Extended Release Tablets before swallowing. Enablex (Darifenacin) Extended Release Tablets may be taken on an empty stomach or with food.
You should take Enablex (Darifenacin) Extended Release Tablets with a full glass of water.
Store Enablex (Darifenacin) Extended Release Tablets at room temperature at 77 degrees F (25 degrees C) in a tightly closed container away from heat and light. Brief storage between 59 and 86 degrees (15 and 30 degrees C) is permitted. Do not store Enablex (Darifenacin) Extended Release Tablets in the bathroom.
Take Enablex (Darifenacin) Extended Release Tablets regularly to receive the most benefit from it. Taking Enablex (Darifenacin) Extended Release Tablets at the same time each day will help you to remember.
Do not miss any doses of Enablex (Darifenacin) Extended Release Tablets. If you miss a dose of Enablex (Darifenacin) Extended Release Tablets, take it as soon as possible. If it is almost time for your next dose, skip the missed dose and go back to your regular dosing schedule. DO NOT take 2 doses of Enablex (Darifenacin) Extended Release Tablets at once.
