药理作用
本品为第2代磺脲类降血糖药,并有降脂和抗凝血作用。其降血糖作用迅速而强,约为甲苯磺丁脲的1000倍,疗效近似格列苯脲,作用机制是刺激胰岛的β细胞,促使胰岛素分泌量增加,同时亦刺激胰岛α细胞使高血糖分泌受抑制。其作用方式和氯磺丙脲相似而毒副作用更低且无致畸现象。此外,有人认为本品尚有抑制肝糖原的分解,促进肌肉利用和消耗葡萄糖的作用;还可能通过胰腺外的作用改变胰岛素靶组织对胰岛素的反应性,增强胰岛素作用,所以有些患者服用单一剂量就可有效地控制血糖达24h。另有文献报道,本品能纠正早期糖尿病的微细血管循环障碍,降低血浆的三酰甘油水平,加快体内胆固醇向HDL的转化,还可以降低血小板的凝聚力和提高对纤维蛋白分解、吸收能力,因而可抵制血小板凝集,减缓毛细血管内血栓形成与内皮损伤,对微血管病变可能有一定的防治作用。利用控释技术,使药物缓慢释放,从而达到持久降血糖作用,药效可维持24h,能降低空腹血糖和餐后血糖。
适应症
用于胰岛素分泌不足的各型糖尿病,包括单用饮食治疗未能达到良好控制的轻、中度2型糖尿病患者及伴有血糖病变的糖尿病。
用法现与用量
通常情况下,每日剂量125〜250毫克口服给药,并确定该维持剂量适当地根据需要增加。然而,每日最大剂量是到500毫克。
施用方法,在一次性给药的情况下早餐前或后,在两剂之前或之后口服早晨和傍晚,分别的情况下。

Effects of this medicine
This medicine acts on the pancreas to stimulate insulin secretion, and consequently lowers blood glucose.
It is usually used in the treatment of insulin-independent diabetes mellitus.
Before using this medicine, be sure to tell your doctor and pharmacist
If you ever experienced any allergic reaction (itch, rash etc.) to any medicine.
If you are a patient with ketosis, diabetic coma or precoma, insulin-dependent diabetes mellitus, hepatic or renal disorder, infections, an injury, diarrhea, or gastrointestinal disorder.
If you are before or after undergoing a surgery.
If you are pregnant or breastfeeding.
If you are taking any other medicinal products. (Some medicines may interact to enhance or diminish medicinal effects. Beware of over-the-counter medicines and dietary supplements as well as other prescription medicines.)
Dosing schedule (How to take this medicine)
Your dosing schedule prescribed by your doctor is <<to be written by a healthcare professional>>
General dosage regimen: For adults, take a half to one tablet (125-250 mg of the active ingredient) daily. The dosage may be increased as necessary and the maintenance dose will be determined. However, the maximum daily dose is limited to 2 tablets (500 mg). If you instructed to take the medicine once daily, take it before or after breakfast, or twice daily, before or after breakfast and supper. Strictly follow the instructions of your doctor/pharmacist.
Skip the missed dose and continue your regular dosing schedule. DO NOT take double doses to make up for the missed dose.
If you took too much of the medicine (more than ordered), check with your doctor/pharmacist.
Do not stop taking the medicine without the instructions of your doctor.
Precautions while taking this medicine
Strictly follow diet and exercise therapy instructed by your doctor.
The medicine may cause hypoglycemia. Pay close attention to avoid any accident when you work at heights, drive a car or operate dangerous machinery after taking the medicine.
If you noticed any symptoms of hypoglycemia (sweating, hungry feeling, shaky limbs, etc.), take sugar or juice including glucose and consult with your doctor. To take a measure for these symptoms, carry sugar with you.
Tell your family members that you take the medicine in case that impairment of consciousness (one of the symptoms of hypoglycemia) occurs.
Possible adverse reactions to this medicine
Common side effects are reported as below. If any of them occurs, check with your doctor/pharmacist: hypoglycemia, loss of appetite, abdominal discomfort, etc.
The symptoms described below are rarely seen as initial symptoms of the adverse reactions indicated in brackets. If any of these symptoms occur, stop taking this medicine and see your doctor immediately.
sweating, hungry feeling, shaky limbs [hypoglycemia]
symptom of anemia, bleeding tendency, sore throat [aplastic anemia, agranulocytosis]

Storage conditions and other information
Keep the medicine out of the reach of children. Store it away from direct sunlight, heat and moisture.
Do not store the remainder and ask your pharmacist how to discard it.
