BALB/c裸鼠脾内移植人源性肝细胞HepLL的生物学特性

【摘要】 目的 研究人源性肝细胞HepLL脾内途径移植后在体内的生物学特性。方法 将人源性肝细胞HepLL移植入切除肝左叶和未切除肝左叶BALB/c裸鼠脾内,通过病理和免疫组化检测其生物学特性。结果 新建的人源性肝细胞移植入BALB/c裸鼠脾脏后可以长期成团存活、增殖并表达肝细胞特异性标记HEP,有糖原的堆积,始终未检测到AFP的表达。至移植后30天,脾脏内移植的HepLL细胞切除左肝组比未切左肝组增生活跃。移植入脾脏的人源性肝细胞可向宿主肝脏移行并进入肝实质,同时可长时间在宿主肝脏内增殖。结论 移植入脾脏的人源性肝细胞HepLL能保持其原有的生物学特性并能移行、整合入肝脏。肝左叶切除造成的肝细胞再生微环境有利于移植的人源性肝细胞HepLL增殖。

Biological characteristics of immortalized human hepatocyte (HepLL) after transplantation in spleen of BALB/c naked mice

【Abstract】 Objective To investigate the biological characteristics of immortalized human hepatocyte (HepLL) after transplantation in the spleen of BALB/c naked mice. Method The HepLL cells were transplanted into the spleen of BALB/c naked mouse and were characterized by the methods of pathology and immunohistochemistry in vivo. Results The human-derived hepatocytes could survive long after transplantation. They proliferated and expressed HEP, a kind of specific marker of hepatocytes,restored glycogen, without the expression of AFP. After transplantation of days 30, the prolification ability of HepLL transplanted in spleen is stronger in group which left liver lobe was resected then in group which it wasn’t resected.The hepatocytes, which were transplanted into the spleen, could migrate and be integrated into the host liver, and then proliferated in the liver for a long time. Conclusion HepLL cells transplanted into spleen can maintain their biological characteristics in vivo while they migrate into the parenchyma of host liver. Resection of the left liver lobe created beneficial microenvironment for the proliferation of human-derived hepatocytes.
